Radiopharmaceutics & PET Radiochemistry MSc – Research Project Titles 1. Effect of COX-2 inhibition on choline uptake and metabolism in prostate cancer

ثبت نشده
چکیده

Abnormal choline metabolism serves as a marker associated with oncogenesis and tumour progression. 11Cand 18F-labeled choline are positron emission tomography (PET) tracers used to study choline uptake. Nuclear magnetic resonance spectroscopy (NMRS) can detect choline and choline metabolites non-invasively; increased levels of these compounds serve as biomarkers of transformation, staging and response to therapy. Furthermore, enzymes of choline metabolism, such as choline kinase, present novel targets for image-guided cancer therapy. The arachidonic acid pathway plays a major role in the development and progression of a number of cancers, including prostate cancer. Epidemiological evidence suggests that nonsteroidal anti-inflammatory drugs may decrease the risk of prostate cancer, possibly through the inhibition of cyclooxygenase-2 (COX-2). Furthermore, COX-2 has been shown to regulate choline phospholipid metabolism, through effects on choline kinase expression, in breast cancer cells. However, studies have not been carried out on the effects of COX-2 on choline metabolism in prostate cancer cells. The project will use 11C (or 18F)-choline to study choline uptake, NMRS to study choline metabolism, and immunoblotting to determine expression of proteins involved in choline uptake and metabolism, in 3 prostate cancer cell lines: androgen-dependent LNCaP cells and androgen-resistant DU145 and PC3 cells which have low, moderate, and high metastatic potential, respectively. The aim of the proposed study is to determine if the degree of cancer aggressiveness correlates with choline uptake and/or metabolism, and if so, whether cancer aggressiveness is affected by COX-2 inhibition. All cell lines will be provided by Dr. Jonathan Morris (Cancer Studies, KCL). Key techniques learned/performed by student: Cell culture, NMR spectroscopy, immunoblotting, cellular PET tracer uptake Student description: This project would suit a biomedical student who may be interested in going on to a Ph.D.

Physiological distribution and range of normal SUVmax values of 18F-Choline PET/CT

Introduction: 18F-Choline PET-CT is an increasingly used technique in patients with prostate cancer. The main indication is to localise the disease in patients with biochemical recurrence. To accurately interpret 18F-Choline PET, knowledge of normal tracer distribution is paramount. The aim of this study was to describe the normal distribution pattern of 18F-Cho...

Diagnostic uncertainty of isolated skull lesion in prostate cancer- Role of SPECT/CT

Skeletal involvement is the second most common site of metastases after lymph nodal metastases in patients with prostate cancer. The skeletal metastases from prostate cancer are osteoblastic in nature and show increased tracer avidity on the bone scan. Focal tracer avid lesion in skeleton especially in skull requires the careful examination by further investigation. The patients with skull meta...

FDA approves 11C-choline for PET in prostate cancer.

T he U.S. Food and Drug Administration (FDA) announced on September 12 approval of production and use of 11C-choline for PET imaging in recurrent prostate cancer. The Mayo Clinic PET Radiochemistry Facility (Rochester, MN) will be the first approved facility to produce the tracer injection. 11C-choline must be produced in a specialized facility and must be administered to patients shortly after...

Evaluation of androgen-induced effects on the uptake of [18F]FDG, [11C]choline and [11C]acetate in an androgen-sensitive and androgen-independent prostate cancer xenograft model

BACKGROUND Androgen deprivation (AD) is generally used as a first-line palliative treatment in prostate cancer (PCa) patients with rising prostate-specific antigen (PSA) after primary therapy. To acquire an accurate detection of tumour viability following AD with positron emission tomography (PET), an androgen-independent uptake of tracers would be advantageous. Several metabolic PET tracers ar...

Synthesis and evaluation of 18F-labeled choline as an oncologic tracer for positron emission tomography: initial findings in prostate cancer.

The up-regulation of rates of choline uptake and phosphorylation in certain malignancies has motivated the development of positron-labeled choline analogues for noninvasive detection of cancer using positron emission tomography (PET). The choline analogue, no-carrier-added [18F]fluoromethyl-dimethyl-2-hydroxyethyl-ammonium (FCH), was synthesized through the intermediate [18F]fluorobromomethane....
